Expression of genes involved in DNA repair and cell cycle checkpoint pathways in Triple Negative compared to Luminal A Breast Cancer: a molecular characterization

چکیده
Purpose Considering the clear emerging role of the DNA repair and the cell cycle checkpoints as predictive, prognostic and therapeutic targets in cancer there is a need to better characterized human tumours to define sub-sets of patients that would benefit of a particular treatment modality. The aim of the present work is to characterize molecularly a cohort of Triple Negative Breast Cancers (TNBC; estrogen receptor negative, progesterone receptor negative, and HER2/neu negative by immunohistochemistry) compared to Luminal A Breast Cancer (LABC; estrogen receptor positive and/or progesterone receptor positive, and HER2/neu negative, ki67 expression < 15% by immunohistochemistry) as regard of gene expression involved in DNA repair pathway (in particular genes involved in nucleotide excision repair, base excision repair, homologous recombination repair and BRCA/Fanconi anemia pathway) and cell cycle checkpoint pathway (CHK1) and their correlation with the clinical-pathological characteristics.

Inherent radiosensitivity and its impact on breast cancer chemo-radiotherapy
About 10% of apparently normal individuals are sensitive to clastogenic effects of physico-chemical agents. More than 45% of breast cancer patients’ exhibit elevated radiosensitivity. Although the nature of inherent radiosensitivity is not fully understood, but insufficiency and impaired DNA repair mechanism might be prime cause of radiosensitivity. This is evident from genetically affect...
متن کاملComparison of BRCA1 Expression between Triple-Negative and Luminal Breast Tumors
Background: Previous studies have suggested that BRCA1 dysregulation has been shown to have a role in triple-negative phenotypic manifestation. However, differences of BRCA1 expression, as a tumor suppressor gene, have rarely been investigated between luminal and triple-negative breast tumors. Therefore, the present study attempted to compare the BRCA1 expression in triple-negative with lu...
متن کاملTriple Negative Breast Cancers Have a Reduced Expression of DNA Repair Genes
DNA repair is a key determinant in the cellular response to therapy and tumor repair status could play an important role in tailoring patient therapy. Our goal was to evaluate the mRNA of 13 genes involved in different DNA repair pathways (base excision, nucleotide excision, homologous recombination, and Fanconi anemia) in paraffin embedded samples of triple negative breast cancer (TNBC) compar...

Background and Aim: Today, cancer is considered as a major health problem and affects the health of society. Breast cancer is the second leading cause of cancer death in women after lung cancer. According to epidemiological studies, cancer is the second most common cause of death after cardiovascular disease worldwide and the third leading cause of death after cardiovascular disease and acciden...

Stable molecular changes during cell division without any change in the sequence of DNA molecules is known as epigenetic. Molecular mechanisms involved in this process, including histone modifications, methylation of DNA, protein complex and RNA antisense. Cancer genome changes happen through a combination of DNA hypermethylation, long-term epigenetic silencing with heterozygosis loss and genom...
